NGF Overview

Nerve growth factor (NGF) is a neuropeptide involved in regulating the growth, proliferation and survival of certain target neurons. When expressed, NGF initially exists as a 7S complex with a molecular weight of 130kDa consisting of three proteins, α-NGF, β-NGF, and γ-NGF(2:1:2 ratio). The term “nerve growth factor” usually refers to the 2.5S beta subunit protein with a molecular weight of 26kDa. The beta subunit is the only biologically active component of the 7SNGF complex (e.g. as a signaling molecule). NGF plays an important role in both innate and acquired immunity. Studies have shown that NGF circulates through plasma and is important for overall homeostasis maintenance. NGF promotes myelin repair and is also involved in a variety of psychiatric disorders, such as Alzheimer’s disease, depression, schizophrenia, autism, Rett syndrome, anorexia nervosa, bulimia nervosa. Dysregulation of NGF signaling is associated with Alzheimer’s disease. Similarly, NGF plays a role in many cardiovascular diseases, such as coronary atherosclerosis, obesity, type 2 diabetes, metabolic syndrome. NGF also promotes wound healing.
